一 、單胞藻純培養室操作描述

儀器使用

(1) 超凈工(gong)作臺

使(shi)用前提前開機(ji),打開紫外線(xian)燈和風機(ji)開關20分鐘。超(chao)凈臺面(mian)上不(bu)存放(fang)不(bu)必(bi)要(yao)的物品,保(bao)持工作(zuo)(zuo)(zuo)區內的凈化氣(qi)流(liu)不(bu)受(shou)干擾(rao),工作(zuo)(zuo)(zuo)時(shi)盡(jin)量避免作(zuo)(zuo)(zuo)明顯(xian)干擾(rao)氣(qi)流(liu)的動作(zuo)(zuo)(zuo);超(chao)凈臺保(bao)持光滑(hua)水平,光滑(hua)便于用酒精(jing)消毒(du)(du)布(bu)擦(ca),水平是(shi)倒培養(yang)基時(shi)使(shi)培養(yang)皿內平板厚度一致(zhi)。工作(zuo)(zuo)(zuo)時(shi)超(chao)凈臺周圍空(kong)氣(qi)流(liu)動要(yao)緩慢,雜菌(jun)(jun)盡(jin)量減(jian)(jian)少(shao),所以(yi)要(yao)定期(qi)清潔(jie)消毒(du)(du)保(bao)種室(shi)內,關閉門窗,盡(jin)可能減(jian)(jian)少(shao)雜菌(jun)(jun)數量;空(kong)氣(qi)過(guo)濾網定期(qi)清潔(jie)。

(2)真(zhen)空泵和溶劑(ji)過(guo)濾器及(ji)過(guo)濾膜(mo)

此環節(jie)過濾只是(shi)為單胞藻純培養室(保種,提純,擴培試管(guan)級(ji))準備清潔無菌海水。其他級(ji)別(bie)的培養無需(xu)此環節(jie)。

過濾(lv)(lv)(lv)膜使(shi)用(yong)0.2微米孔徑(jing),不可用(yong)手拿(na),用(yong)鑷子選取。真空(kong)泵(beng)與溶劑過濾(lv)(lv)(lv)器(qi)之間通過合適口(kou)徑(jing)的橡膠(jiao)管連接緊(jin)密(mi),每次(ci)過濾(lv)(lv)(lv)要(yao)注意濾(lv)(lv)(lv)膜的過濾(lv)(lv)(lv)效果,防止濾(lv)(lv)(lv)孔阻塞影響過濾(lv)(lv)(lv)效果。濾(lv)(lv)(lv)瓶內的濾(lv)(lv)(lv)液不能沒過濾(lv)(lv)(lv)板(ban)出水口(kou),否則(ze)濾(lv)(lv)(lv)液將(jiang)被抽吸入真空(kong)泵(beng),導(dao)致(zhi)真空(kong)泵(beng)損壞。

應該使濾板內的營養(yang)液過(guo)濾盡后(無水滴)才能停泵。要定期注(zhu)意真空泵內油(you)量(liang),注(zhu)意加油(you)。

(3) 恒溫鼓風干燥箱

設(she)(she)(she)定(ding)(ding)溫(wen)度時(shi)。先按控(kong)溫(wen)儀功(gong)能(neng)鍵(jian)(jian)(jian)SET進入溫(wen)度設(she)(she)(she)定(ding)(ding)狀態,此時(shi)SP設(she)(she)(she)定(ding)(ding)顯(xian)示(shi)一(yi)閃(shan)(shan)一(yi)閃(shan)(shan),要按移動(dong)位鍵(jian)(jian)(jian)配(pei)合加鍵(jian)(jian)(jian)或減鍵(jian)(jian)(jian),設(she)(she)(she)定(ding)(ding)時(shi)先從分位數字設(she)(she)(she)定(ding)(ding),一(yi)直到百(bai)位數,設(she)(she)(she)定(ding)(ding)結束需按一(yi)下功(gong)能(neng)鍵(jian)(jian)(jian)SET,此時(shi)干燥箱進入升溫(wen)狀態,加熱指(zhi)示(shi)燈亮,當箱內(nei)溫(wen)度接近設(she)(she)(she)定(ding)(ding)溫(wen)度時(shi),加熱指(zhi)示(shi)燈閃(shan)(shan)動(dong)多次,控(kong)制進入恒溫(wen)狀態。

使(shi)用注(zhu)意事項(xiang):物品不能堆得太滿,更不能堵住出風口,以免影響溫度均勻上升。若是有牛皮紙和保鮮膜時,溫度不能超過180度。取出物品時,箱內外溫差不宜過高,防止玻璃器皿炸裂。橡膠制品不能放入。

(4)加濕器

配(pei)合生化培(pei)養(yang)箱使用(yong)。

水源是(shi)蒸餾水;不應容器(qi)內帶水移(yi)動(dong)加(jia)濕(shi)器(qi);加(jia)水時應在容器(qi)上表示的滿(man)水線(xian)為止。

應注意(yi)的事項:平時留意(yi)觀察水(shui)箱的存水(shui)量(liang),及時予以補充,遠離熱源(yuan),腐蝕物。

(5)蒸(zheng)汽消毒(du)器

容(rong)器加(jia)水(shui)時,水(shui)位(wei)一定要超過電(dian)熱(re)(re)管,最(zui)好是(shi)到3個(ge)支撐點的(de)(de)下緣,如果連(lian)續使用(yong),應在每次消毒前補足蒸(zheng)發水(shui)量,不然少水(shui)會(hui)燒壞(huai)電(dian)熱(re)(re)管。蓋上的(de)(de)放(fang)氣軟管插入消毒桶圓槽內,對(dui)正蓋和主體(ti)(ti)的(de)(de)螺栓槽,依次將相對(dui)方的(de)(de)翼型螺母予(yu)以(yi)旋緊(jin),使蓋與主體(ti)(ti)密合(he),加(jia)熱(re)(re)開(kai)始時,應將放(fang)氣閥與安全閥的(de)(de)小手(shou)柄扳(ban)至垂直放(fang)氣方位(wei),必(bi)須待有較(jiao)急(ji)蒸(zheng)汽噴出,即將小手(shou)柄扳(ban)回水(shui)平“關閉”方位(wei)。

當到(dao)達所需壓(ya)力(li)和(he)溫度(du),計算消毒時間,消毒畢后(hou)(hou),關(guan)閉(bi)電源,待壓(ya)力(li)指針指向0時,將放氣(qi)(qi)閥的小手柄扳至垂直放氣(qi)(qi)方位,氣(qi)(qi)放完后(hou)(hou),打開容器蓋。物(wu)品消毒后(hou)(hou)要迅速干燥。

應注意滅(mie)菌液體(ti)時(shi),不(bu)(bu)超過(guo)盛(sheng)液體(ti)容器體(ti)積的(de)(de)3/4,瓶(ping)塞(sai)(sai)不(bu)(bu)可用軟木塞(sai)(sai)或未打孔(kong)的(de)(de)橡膠塞(sai)(sai)。滅(mie)菌器內冷(leng)空(kong)(kong)氣(qi)必(bi)須完全排除(chu)。因為空(kong)(kong)氣(qi)的(de)(de)膨脹壓(ya)(ya)(ya)(ya)(ya)大(da)于水(shui)蒸(zheng)氣(qi)的(de)(de)膨脹壓(ya)(ya)(ya)(ya)(ya),當水(shui)蒸(zheng)氣(qi)含(han)冷(leng)空(kong)(kong)氣(qi)時(shi),壓(ya)(ya)(ya)(ya)(ya)力表所表示的(de)(de)壓(ya)(ya)(ya)(ya)(ya)力是水(shui)蒸(zheng)氣(qi)壓(ya)(ya)(ya)(ya)(ya)力和部分冷(leng)空(kong)(kong)氣(qi)壓(ya)(ya)(ya)(ya)(ya)力之和,不(bu)(bu)是水(shui)蒸(zheng)氣(qi)的(de)(de)實(shi)際(ji)壓(ya)(ya)(ya)(ya)(ya)力,而同一(yi)壓(ya)(ya)(ya)(ya)(ya)力下的(de)(de)實(shi)際(ji)溫(wen)度,含(han)空(kong)(kong)氣(qi)的(de)(de)蒸(zheng)汽低(di)于飽和蒸(zheng)汽,這(zhe)樣導致達(da)不(bu)(bu)了(le)消(xiao)毒(du)效果。絕(jue)不(bu)(bu)可堵塞(sai)(sai)安(an)全閥出氣(qi)孔(kong),必(bi)須留空(kong)(kong)位(wei)保證其暢通(tong)放氣(qi),發現超溫(wen)超壓(ya)(ya)(ya)(ya)(ya),立即斷電。

蒸餾器

先關(guan)閉(bi)放(fang)水(shui)(shui)(shui)(shui)(shui)閥(fa),打開進水(shui)(shui)(shui)(shui)(shui)閥(fa),待(dai)溢(yi)水(shui)(shui)(shui)(shui)(shui)皮(pi)管(guan)有水(shui)(shui)(shui)(shui)(shui)流(liu)出(chu),方可(ke)接通電源,此(ci)時可(ke)暫(zan)時關(guan)閉(bi)進水(shui)(shui)(shui)(shui)(shui)閥(fa),待(dai)出(chu)水(shui)(shui)(shui)(shui)(shui)皮(pi)管(guan)有蒸(zheng)(zheng)餾(liu)(liu)水(shui)(shui)(shui)(shui)(shui)流(liu)出(chu),打開進水(shui)(shui)(shui)(shui)(shui)閥(fa),制備蒸(zheng)(zheng)餾(liu)(liu)水(shui)(shui)(shui)(shui)(shui)。定期清洗蒸(zheng)(zheng)餾(liu)(liu)器內壁,除去水(shui)(shui)(shui)(shui)(shui)垢(gou),不(bu)然影(ying)響蒸(zheng)(zheng)餾(liu)(liu)水(shui)(shui)(shui)(shui)(shui)質量和使(shi)用效果。切(qie)不(bu)可(ke)關(guan)閉(bi)水(shui)(shui)(shui)(shui)(shui)閥(fa)制備蒸(zheng)(zheng)餾(liu)(liu)水(shui)(shui)(shui)(shui)(shui),否則(ze)燒壞電熱管(guan)。

(6)鼓風機

定期更換(huan)吸氣口的過(guo)濾棉(mian),異常情況,請電工檢查(cha)。

(7)空(kong)調(diao)

保(bao)種室若有(you)生長藻種,需維持恒定溫(wen)度,反之(zhi)關閉。一級,二級空調(diao)視外界氣溫(wen)調(diao)整(zheng)開關。

二 、衛生消毒

工具衛生消毒

1、濾水筒(tong)式(shi)(shi)過濾器(qi)。筒(tong)式(shi)(shi)過濾器(qi)的(de)濾芯應用(yong)一次更換一次,用(yong)過的(de)濾芯浸泡在1%鹽酸溶液中消毒(du)12h以(yi)上。然后(hou)用(yong)洗(xi)潔(jie)精清(qing)(qing)洗(xi),再(zai)用(yong)足量清(qing)(qing)水沖洗(xi)干(gan)凈,晾干(gan)備用(yong)。(濾芯規格應小于等于0.5微米)。

2、試(shi)管(guan)(規格(ge)10毫(hao)升到20毫(hao)升),錐形瓶(50毫(hao)升到200毫(hao)升),500ml錐形瓶,2.5L瓶廣口瓶,移液管(guan),移液槍管(guan)/盒,玻璃氣(qi)(qi)管(guan),膠氣(qi)(qi)管(guan),氣(qi)(qi)石(shi)(shi),溶劑過濾(lv)器,培養皿。用(yong)(yong)(yong)過后應當用(yong)(yong)(yong)清(qing)(qing)水(shui)清(qing)(qing)洗后浸(jin)泡于1% 鹽(yan)酸12h以上(溶劑過濾(lv)器除外(wai),浸(jin)泡時(shi)相(xiang)近(jin)大(da)小(xiao)器皿分類放不同的消毒桶,規格(ge)相(xiang)差大(da),易(yi)導致相(xiang)互碰(peng)撞碎爛)然后用(yong)(yong)(yong)洗潔精清(qing)(qing)洗(用(yong)(yong)(yong)毛刷清(qing)(qing)洗試(shi)管(guan)底部(bu)(bu)時(shi),不可太(tai)用(yong)(yong)(yong)力,否(fou)則底部(bu)(bu)易(yi)破裂(lie)),再用(yong)(yong)(yong)大(da)量清(qing)(qing)水(shui)沖洗干(gan)(gan)凈,風(feng)干(gan)(gan)或(huo)曬干(gan)(gan),高壓滅菌后,放入(ru)恒(heng)溫干(gan)(gan)燥(zao)(zao)箱(膠氣(qi)(qi)管(guan),氣(qi)(qi)石(shi)(shi)除外(wai))干(gan)(gan)燥(zao)(zao)備用(yong)(yong)(yong)。

3、聚乙烯藻(zao)袋用(yong)(yong)(yong)于二(er)級(ji)藻(zao)類(lei)藻(zao)種掛袋培養或3級(ji)擴(kuo)培(一般使用(yong)(yong)(yong)盛裝(zhuang)藻(zao)液(ye)12升(sheng)左右)。先用(yong)(yong)(yong)清水沖(chong)洗(xi)干凈內外,浸泡于1% 鹽酸(suan)12h以(yi)上,再(zai)用(yong)(yong)(yong)大量(liang)清水沖(chong)洗(xi)干凈,風干或曬干,備(bei)用(yong)(yong)(yong)。由(you)于藻(zao)袋近似密封(feng)性(xing),對(dui)于防止微(wei)生物(wu)污染(ran)有很好的效果。

4、毛巾、保種室,一(yi)級和(he)二級毛巾不可混用,所用毛巾后(hou)都(dou)應(ying)消毒清洗(xi)晾干(gan)后(hou)備用。

5、錫(xi)箔紙(zhi):除非粘(zhan)上(shang)油污(wu)或(huo)污(wu)物以及破損(sun),都(dou)應重復使用(yong)。

6、牛(niu)皮紙:定期拿(na)到(dao)烘箱(xiang)烘干。

7、定(ding)期更(geng)換空氣(qi)(qi)過濾器(用于1,2級(ji)培養的進氣(qi)(qi)過濾)的濾芯,濾芯已油化(hua)或沖(chong)散的棄去。

8、定(ding)期更換鼓風(feng)機進風(feng)口的過濾棉。

9、對新購入(ru)的器皿應(ying)先除去包裝沾(zhan)染(ran)的污垢后,用(yong)(yong)洗(xi)潔精刷洗(xi),流(liu)水沖(chong)凈,再(zai)浸泡于1%的鹽酸12h,使游離的堿性(xing)物(wu)質(zhi)除去,再(zai)用(yong)(yong)清水沖(chong)洗(xi)干凈,風干或曬干備用(yong)(yong)。

室內衛生消毒

1、保種室每(mei)天早上用清水擦洗實驗臺(tai)面(mian),地板,墻壁,木架后,開紫(zi)外燈(deng)消毒20分鐘,此(ci)時關(guan)掉(diao)空(kong)調,關(guan)掉(diao)日光燈(deng),由于(yu)紫(zi)外燈(deng)照射會產生一(yi)定(ding)量的臭氧,對(dui)人體(ti)有害,所(suo)以消毒關(guan)閉紫(zi)外燈(deng)后人應隔一(yi)段時間(jian)再進去。進保種室換(huan)鞋。

2、一級和二級室每(mei)天清潔,接(jie)種前接(jie)種后承載(zai)物框架都應(ying)用(yong)消毒(du)液(ye)消毒(du)。

3、門(men)口消毒池定期更(geng)換消毒藥水(高錳酸鉀)。

4、常用消(xiao)毒劑濃度 1%鹽酸,次氯酸鈉20ppm(有效氯),碘液(1000ppm),酒精70-75%,高錳酸鉀(jia)1mg/L。

三 、接種

藻種固體培養基制備

所需燒杯(bei),玻(bo)棒須(xu)(xu)(xu)清(qing)洗干凈,錐(zhui)形瓶(ping)和培(pei)養(yang)(yang)皿須(xu)(xu)(xu)消毒清(qing)洗滅(mie)菌(jun)烘干。將(jiang)稱好的(de)純(chun)化瓊(qiong)(qiong)(qiong)脂粉加入(ru)燒杯(bei)再加入(ru)一定量的(de)海水(shui)(shui),加熱(re)(必須(xu)(xu)(xu)不停攪拌(ban),以(yi)免瓊(qiong)(qiong)(qiong)脂糊底燒焦(jiao))至完全融化,加熱(re)后(hou)的(de)瓊(qiong)(qiong)(qiong)脂培(pei)養(yang)(yang)基應當(dang)透明無顯(xian)著沉(chen)淀,若(ruo)明顯(xian),可采用(yong)清(qing)潔的(de)白色薄(bo)絨布或中間夾有薄(bo)層(ceng)吸水(shui)(shui)棉(mian)的(de)雙層(ceng)紗布鋪在漏斗上(shang)趁熱(re)過濾,融化畢后(hou)精確(que)加入(ru)營養(yang)(yang)鹽(維生素B溶液須(xu)(xu)(xu)等培(pei)養(yang)(yang)基高壓(ya)滅(mie)菌(jun)后(hou)冷卻(que)至50攝氏度(du)以(yi)下再加)。

培(pei)(pei)養基采(cai)用121攝(she)氏度(du)高(gao)壓蒸汽滅菌20至(zhi)30分鐘即可,培(pei)(pei)養基滅菌后(hou)應立即取出,冷(leng)卻至(zhi)50攝(she)氏度(du)以(yi)下(xia),即加維(wei)生素B溶(rong)液。將盛裝培(pei)(pei)養基的錐(zhui)形(xing)瓶(ping)和培(pei)(pei)養皿放(fang)(fang)在超凈工作臺上(shang),點燃酒(jiu)精燈,右手托起錐(zhui)形(xing)瓶(ping)底(di)(di),將瓶(ping)口在酒(jiu)精燈稍加燒(shao),然后(hou)迅速(su)將培(pei)(pei)養基倒入培(pei)(pei)養皿中,以(yi)鋪滿(man)皿底(di)(di)為止(厚(hou)薄適中)。鋪放(fang)(fang)好后(hou),放(fang)(fang)置20-30分鐘,待(dai)培(pei)(pei)養基凝固后(hou),即開始操(cao)作。

若是(shi)做斜面培(pei)養基(ji)(ji),則加入維(wei)生素B溶(rong)液后(hou)(hou),將盛裝(zhuang)培(pei)養基(ji)(ji)的(de)錐形(xing)瓶和試(shi)管以及一根厚約1厘米的(de)小木板放(fang)在超凈工(gong)作臺上。分裝(zhuang)約為試(shi)管高度的(de)1/3-2/3(視情況操(cao)作),分裝(zhuang)量應以能(neng)形(xing)成(cheng)2/3底層和1/3斜面量為好(hao),擺置成(cheng)適當斜面,待(dai)其(qi)自然(ran)凝固。培(pei)養基(ji)(ji)制備好(hao)后(hou)(hou),如(ru)發現破裂,水分浸入,色澤(ze)異常(chang)等均應棄去.

待凝固后(hou)即進行(xing)操作(zuo),現一般采(cai)用平(ping)板劃(hua)線(xian)接(jie)種(zhong)和涂布接(jie)種(zhong)。

涂布接種

所(suo)用種液必須經(jing)過弧菌檢測(ce)合格(ge)后方可(ke)使用。涂(tu)布棒(bang)在火(huo)(huo)焰上慢慢來回過火(huo)(huo)焰3次,冷卻(不(bu)可(ke)驟冷),將(jiang)種液用移液槍(qiang)從試管(guan)吸出注入(ru)平板上面(mian)(槍(qiang)管(guan)不(bu)可(ke)碰及試管(guan)外壁,更(geng)不(bu)能碰及其(qi)它(ta)物品或臺面(mian)),迅(xun)速(su)用涂(tu)布棒(bang)在表面(mian)作(zuo)來回左右的涂(tu)布,讓藻液分布均勻,畢后,封(feng)口(kou)同平板劃(hua)線。

試管液體培養基接種

所用種液必須經過弧菌(jun)(jun)(jun)檢(jian)測合格(ge)后方可使用。火(huo)焰滅(mie)菌(jun)(jun)(jun)前試(shi)管(guan)(guan)(guan)口(kou)外(wai)壁不(bu)能有(you)水滴(易導致受熱不(bu)均(jun)而破(po)裂(lie)),燒試(shi)管(guan)(guan)(guan)口(kou)時(shi)(shi),試(shi)管(guan)(guan)(guan)傾斜(xie)45度角(jiao),不(bu)斷移(yi)動試(shi)管(guan)(guan)(guan),使試(shi)管(guan)(guan)(guan)口(kou)受熱均(jun)勻,滅(mie)菌(jun)(jun)(jun)時(shi)(shi)間3秒左右。滅(mie)菌(jun)(jun)(jun)后不(bu)能驟冷,防止(zhi)試(shi)管(guan)(guan)(guan)破(po)裂(lie)。畢后,用盛裝營養液150或200ml的錐形瓶(ping)(也(ye)要滅(mie)菌(jun)(jun)(jun)瓶(ping)口(kou))向試(shi)管(guan)(guan)(guan)中傾倒(dao)營養液,每(mei)次(ci)只能拿一(yi)個試(shi)管(guan)(guan)(guan)(注(zhu):若幾個試(shi)管(guan)(guan)(guan)對比時(shi)(shi),可以同時(shi)(shi)拿在(zai)手里,用大拇指和(he)手掌),倒(dao)好后,同樣試(shi)管(guan)(guan)(guan)和(he)錐形瓶(ping)口(kou)滅(mie)菌(jun)(jun)(jun),試(shi)管(guan)(guan)(guan)塞也(ye)要滅(mie)菌(jun)(jun)(jun),塞試(shi)管(guan)(guan)(guan)塞時(shi)(shi)不(bu)要移(yi)動試(shi)管(guan)(guan)(guan)迎接塞,以免試(shi)管(guan)(guan)(guan)在(zai)移(yi)動中納入可能含(han)菌(jun)(jun)(jun)空氣,火(huo)焰旁為無菌(jun)(jun)(jun)。

所(suo)有試管(guan)(guan)分(fen)裝好后,即(ji)接(jie)種(zhong)。用移(yi)液槍(qiang)選(xuan)(xuan)取(qu)(qu)移(yi)液量(試管(guan)(guan)口滅(mie)菌),注入新(xin)培養(yang)(yang)基(ji),槍(qiang)管(guan)(guan)不(bu)可碰及(ji)管(guan)(guan)外壁,更(geng)不(bu)能碰及(ji)其它物品或臺面,一(yi)(yi)(yi)支槍(qiang)管(guan)(guan)只能和(he)同一(yi)(yi)(yi)種(zhong)液使用,選(xuan)(xuan)取(qu)(qu)另一(yi)(yi)(yi)試管(guan)(guan)種(zhong)液時必須更(geng)換(huan)槍(qiang)頭。接(jie)種(zhong)畢(bi)后,選(xuan)(xuan)取(qu)(qu)合適(shi)光照(zhao)溫度培養(yang)(yang)。由于試管(guan)(guan)培養(yang)(yang)是靜置培養(yang)(yang),而在靜置條件下,藻細胞(bao)會聚(ju)集在試管(guan)(guan)底部,且(qie)藻細胞(bao)周圍會形成透過養(yang)(yang)分(fen)的(de)(de)屏障,營(ying)養(yang)(yang)鹽(yan)的(de)(de)攝入受(shou)阻,光照(zhao)吸收不(bu)均,為使營(ying)養(yang)(yang)鹽(yan)攝入正(zheng)常,藻細胞(bao)受(shou)光均勻,需要(yao)定時搖置試管(guan)(guan)(手先噴消毒酒精,手腕使勁搖,不(bu)要(yao)擺臂(bei),不(bu)能倒置搖)

三角瓶,500ml瓶,2.5L瓶液體培養基接種

三角(jiao)瓶(ping)接(jie)種(zhong)需(xu)無菌操(cao)作。500ml瓶(ping),2.5L瓶(ping)液體培(pei)養(yang)(yang)基接(jie)種(zhong)均(jun)(jun)在室內衛生條(tiao)件下操(cao)作。3種(zhong)瓶(ping)和所需(xu)營(ying)養(yang)(yang)液及(ji)錫(xi)紙,移液管(guan),洗(xi)耳(er)球,氣管(guan),氣石均(jun)(jun)需(xu)經消毒清洗(xi),滅(mie)菌后使用(yong)。三角(jiao)瓶(ping)需(xu)定時搖置,500ml,2.5L瓶(ping)培(pei)養(yang)(yang)在通氣條(tiao)件下培(pei)養(yang)(yang)。

二級聚乙烯藻袋液體培養基接種

氣管,氣石(shi)均(jun)需經(jing)消毒清洗(xi),滅菌(jun)后(hou)使(shi)用(yong),抽(chou)藻(zao)管前(qian)次用(yong)畢后(hou)應(ying)(ying)當(dang)用(yong)水(shui)(shui)泵將(jiang)管內灌(guan)滿消毒鹽酸溶液(ye),今次用(yong)前(qian)需用(yong)海水(shui)(shui)或淡水(shui)(shui)沖洗(xi)干凈(jing)后(hou)使(shi)用(yong)。選取的(de)(de)種液(ye)應(ying)(ying)當(dang)是無明顯沉(chen)淀,泡(pao)沫(mo),原生動(dong)物(wu),附著物(wu),色澤正(zheng)常,藻(zao)細(xi)胞密度大,形態正(zheng)常,弧菌(jun)檢測合格后(hou)選用(yong)。所用(yong)海水(shui)(shui)漂(piao)(piao)白(bai)水(shui)(shui)消毒有效氯應(ying)(ying)足10ppm,曝氣12小(xiao)時(shi),檢測余氯,加硫(liu)代硫(liu)酸鈉中和,方可使(shi)用(yong)。若使(shi)用(yong)稀釋(shi)的(de)(de)漂(piao)(piao)白(bai)水(shui)(shui),應(ying)(ying)當(dang)天配(pei)制當(dang)天使(shi)用(yong),而未使(shi)用(yong)的(de)(de)要在(zai)24小(xiao)時(shi)后(hou)棄去。且稀釋(shi)的(de)(de)漂(piao)(piao)白(bai)水(shui)(shui)必須加蓋放(fang)在(zai)避光的(de)(de)容器內保存。由于漂(piao)(piao)白(bai)水(shui)(shui)會隨(sui)時(shi)間漸漸分(fen)解,一次性購(gou)入的(de)(de)漂(piao)(piao)白(bai)水(shui)(shui)不應(ying)(ying)太多,不要過量(liang)儲(chu)存,以免(mian)影響(xiang)滅菌(jun)功(gong)效。

1、母液(ye)(ye)的(de)配置(zhi)可以參考教(jiao)科(ke)書上所寫,然(ran)后(hou)結合當地的(de)水質(zhi)適(shi)當修改即可。氮,磷(lin),鐵元素,維生素B1,B12是必須使用,維生素母液(ye)(ye)保存于低溫弱光環(huan)境(jing)下(xia),其中維生素的(de)添加(jia)需要先高壓滅菌(jun)后(hou)再加(jia)入(ru)母液(ye)(ye)中。其余營養(yang)鹽(yan)先配置(zhi)好后(hou),再滅菌(jun)。如(ru)果(guo)是硅藻類,還應加(jia)上硅酸(suan)鈉。

4、保(bao)種培養(yang)基(ji)制備:按1.5%的比(bi)例加瓊脂粉(fen),煮沸后加營養(yang)鹽(母液(ye)千(qian)分(fen)之1添加),高(gao)壓(ya)(ya)滅菌。維生素(su)需(xu)高(gao)壓(ya)(ya)滅菌后培養(yang)基(ji)冷(leng)卻至(zhi)30攝氏度方可加入。(可用劃線法(fa),涂布法(fa)等分(fen)離培養(yang)藻(zao)種)。

三級藻生產

三(san)級藻系統包括用水處(chu)理以及(ji)配(pei)送(song),溫(wen)度與光照控制,藻種的供應(ying)等。為確保擴大培(pei)(pei)養(yang)成功,從接種到收(shou)獲的培(pei)(pei)養(yang)時間應(ying)該盡量(liang)縮短,以避(bi)免因為開(kai)放式培(pei)(pei)養(yang)隨(sui)培(pei)(pei)養(yang)規模擴大接觸外界感染源時間加長,三(san)級藻池滋生過多的有害生物(wu),使三(san)級培(pei)(pei)養(yang)失敗。
