對蝦(xia)種苗(miao)(miao)(miao)(miao)(miao)企業(ye)的(de)(de)(de)生存(cun)主要是(shi)依靠(kao)蝦(xia)苗(miao)(miao)(miao)(miao)(miao)養(yang)殖效果來決定的(de)(de)(de),蝦(xia)苗(miao)(miao)(miao)(miao)(miao)成活(huo)率的(de)(de)(de)高低(di)直(zhi)接影響著產業(ye)鏈(lian)的(de)(de)(de)上上下(xia)下(xia)。單(dan)(dan)胞(bao)藻(zao)(zao)對于蝦(xia)苗(miao)(miao)(miao)(miao)(miao)早期是(shi)最適合(he)的(de)(de)(de)餌料,如果培育(yu)(yu)(yu)得當(dang),不(bu)僅可以(yi)節約成本,減少育(yu)(yu)(yu)苗(miao)(miao)(miao)(miao)(miao)水(shui)體環境負荷,尤(you)其是(shi)對于蝦(xia)苗(miao)(miao)(miao)(miao)(miao)的(de)(de)(de)發(fa)育(yu)(yu)(yu)打下(xia)堅(jian)實(shi)的(de)(de)(de)基礎,提高成活(huo)率。結合(he)個(ge)人(ren)的(de)(de)(de)育(yu)(yu)(yu)苗(miao)(miao)(miao)(miao)(miao)感受,投(tou)(tou)喂(wei)單(dan)(dan)胞(bao)藻(zao)(zao)與不(bu)投(tou)(tou)喂(wei)的(de)(de)(de)區別還是(shi)有的(de)(de)(de),比(bi)如苗(miao)(miao)(miao)(miao)(miao)的(de)(de)(de)變態(tai)時(shi)間縮短,活(huo)力(li)個(ge)頭大(da)小,水(shui)質均優于未(wei)投(tou)(tou)喂(wei)單(dan)(dan)胞(bao)藻(zao)(zao)的(de)(de)(de)苗(miao)(miao)(miao)(miao)(miao),當(dang)然如果培育(yu)(yu)(yu)不(bu)得當(dang),反作用(yong)也有,比(bi)如投(tou)(tou)喂(wei)有害菌(jun)超標的(de)(de)(de)藻(zao)(zao)液,藻(zao)(zao)類老化(hua),投(tou)(tou)喂(wei)過量等(deng)。所以(yi)合(he)理(li)運用(yong),方(fang)可收到(dao)理(li)想效果。限于個(ge)人(ren)水(shui)平,描述方(fang)法的(de)(de)(de)錯誤不(bu)足之處,懇請大(da)家指正(zheng)。

一 、單胞藻純培養室操作描述

儀器使用

(1) 超(chao)凈工作臺

使用前提前開(kai)機,打開(kai)紫(zi)外線燈和風(feng)機開(kai)關(guan)20分鐘。超(chao)凈臺(tai)(tai)面上不存放不必要的(de)物(wu)品,保持(chi)工(gong)作(zuo)區內(nei)的(de)凈化氣流(liu)不受(shou)干擾,工(gong)作(zuo)時盡(jin)量避免作(zuo)明(ming)顯干擾氣流(liu)的(de)動(dong)作(zuo);超(chao)凈臺(tai)(tai)保持(chi)光滑水(shui)平,光滑便于用酒精消毒(du)布擦,水(shui)平是倒培養基時使培養皿內(nei)平板厚度一(yi)致。工(gong)作(zuo)時超(chao)凈臺(tai)(tai)周(zhou)圍空氣流(liu)動(dong)要緩慢,雜(za)菌盡(jin)量減少(shao),所以要定期清(qing)潔(jie)消毒(du)保種(zhong)室(shi)內(nei),關(guan)閉門窗,盡(jin)可能減少(shao)雜(za)菌數量;空氣過濾網定期清(qing)潔(jie)。

(2)真(zhen)空泵和溶(rong)劑過濾器及(ji)過濾膜

此(ci)環(huan)節過(guo)濾只是為單胞藻純培(pei)養室(保種,提純,擴培(pei)試管級(ji))準備清潔(jie)無(wu)菌海水。其(qi)他級(ji)別的培(pei)養無(wu)需此(ci)環(huan)節。

過(guo)(guo)濾(lv)(lv)(lv)(lv)(lv)膜使用(yong)0.2微米孔徑,不可用(yong)手拿,用(yong)鑷(nie)子選取。真空(kong)(kong)泵與溶劑過(guo)(guo)濾(lv)(lv)(lv)(lv)(lv)器之(zhi)間(jian)通過(guo)(guo)合(he)適口徑的橡(xiang)膠管連接緊密,每次過(guo)(guo)濾(lv)(lv)(lv)(lv)(lv)要注意濾(lv)(lv)(lv)(lv)(lv)膜的過(guo)(guo)濾(lv)(lv)(lv)(lv)(lv)效果(guo),防(fang)止(zhi)濾(lv)(lv)(lv)(lv)(lv)孔阻塞影(ying)響過(guo)(guo)濾(lv)(lv)(lv)(lv)(lv)效果(guo)。濾(lv)(lv)(lv)(lv)(lv)瓶內的濾(lv)(lv)(lv)(lv)(lv)液不能沒(mei)過(guo)(guo)濾(lv)(lv)(lv)(lv)(lv)板出水口,否則濾(lv)(lv)(lv)(lv)(lv)液將被抽吸入真空(kong)(kong)泵,導(dao)致真空(kong)(kong)泵損壞。

應(ying)該使濾(lv)板內的營養液過濾(lv)盡后(hou)(無水滴)才能停泵。要定期注意(yi)真空(kong)泵內油量,注意(yi)加油。

(3) 恒溫鼓風干燥箱

設定(ding)(ding)溫(wen)度(du)時(shi)。先按控(kong)溫(wen)儀功(gong)(gong)能鍵SET進(jin)入溫(wen)度(du)設定(ding)(ding)狀態,此時(shi)SP設定(ding)(ding)顯示(shi)一(yi)閃一(yi)閃,要(yao)按移(yi)動(dong)位鍵配(pei)合(he)加鍵或減鍵,設定(ding)(ding)時(shi)先從(cong)分(fen)位數字設定(ding)(ding),一(yi)直到百位數,設定(ding)(ding)結束需(xu)按一(yi)下功(gong)(gong)能鍵SET,此時(shi)干燥箱進(jin)入升溫(wen)狀態,加熱指(zhi)示(shi)燈(deng)亮,當(dang)箱內溫(wen)度(du)接近設定(ding)(ding)溫(wen)度(du)時(shi),加熱指(zhi)示(shi)燈(deng)閃動(dong)多次,控(kong)制進(jin)入恒溫(wen)狀態。

使用注意(yi)事項(xiang):物(wu)品不能堆(dui)得太(tai)滿,更(geng)不能堵住出風口,以免影響溫(wen)(wen)度(du)均勻上升。若是有(you)牛皮紙和保鮮膜時(shi),溫(wen)(wen)度(du)不能超過(guo)180度(du)。取(qu)出物(wu)品時(shi),箱(xiang)內外(wai)溫(wen)(wen)差不宜過(guo)高,防止玻璃器皿炸裂。橡膠制品不能放入。

(4)加濕器

配合生化培養箱(xiang)使(shi)用(yong)。

水(shui)(shui)(shui)源(yuan)是蒸餾水(shui)(shui)(shui);不應容(rong)器(qi)內帶水(shui)(shui)(shui)移動加濕器(qi);加水(shui)(shui)(shui)時應在(zai)容(rong)器(qi)上(shang)表示的滿水(shui)(shui)(shui)線為止。

應注意(yi)的(de)事項:平(ping)時(shi)留意(yi)觀察水箱的(de)存水量(liang),及時(shi)予以補(bu)充,遠(yuan)離熱源(yuan),腐蝕物。

(5)蒸(zheng)汽(qi)消毒器

容(rong)器加(jia)水(shui)時(shi),水(shui)位(wei)一定(ding)要(yao)超過電(dian)熱管,最好是到3個支(zhi)撐點的下緣,如果連續使用(yong),應在每次(ci)消(xiao)毒前補足蒸發水(shui)量,不然少水(shui)會燒(shao)壞電(dian)熱管。蓋(gai)上(shang)的放氣(qi)軟管插入消(xiao)毒桶圓槽內,對(dui)正蓋(gai)和主體的螺栓槽,依次(ci)將相對(dui)方的翼型(xing)螺母予以(yi)旋(xuan)緊(jin),使蓋(gai)與(yu)主體密(mi)合,加(jia)熱開始時(shi),應將放氣(qi)閥與(yu)安(an)全閥的小(xiao)手(shou)柄扳至垂直放氣(qi)方位(wei),必須待有較急蒸汽(qi)噴出,即將小(xiao)手(shou)柄扳回水(shui)平“關閉(bi)”方位(wei)。

當到達所需壓(ya)力和(he)溫度,計算消(xiao)毒(du)時間,消(xiao)毒(du)畢后(hou),關閉電源,待壓(ya)力指(zhi)針指(zhi)向0時,將放氣閥(fa)的小手(shou)柄扳至垂直放氣方位,氣放完后(hou),打開(kai)容器蓋。物品消(xiao)毒(du)后(hou)要(yao)迅速干燥。

應注意滅菌液體時(shi),不(bu)超過盛液體容器體積的(de)3/4,瓶塞(sai)不(bu)可用軟木塞(sai)或未打孔的(de)橡膠塞(sai)。滅菌器內冷空(kong)氣(qi)(qi)必(bi)須完(wan)全排(pai)除。因(yin)為空(kong)氣(qi)(qi)的(de)膨(peng)(peng)脹(zhang)壓(ya)(ya)(ya)(ya)大于水(shui)蒸氣(qi)(qi)的(de)膨(peng)(peng)脹(zhang)壓(ya)(ya)(ya)(ya),當水(shui)蒸氣(qi)(qi)含冷空(kong)氣(qi)(qi)時(shi),壓(ya)(ya)(ya)(ya)力表所表示的(de)壓(ya)(ya)(ya)(ya)力是(shi)(shi)水(shui)蒸氣(qi)(qi)壓(ya)(ya)(ya)(ya)力和部分冷空(kong)氣(qi)(qi)壓(ya)(ya)(ya)(ya)力之和,不(bu)是(shi)(shi)水(shui)蒸氣(qi)(qi)的(de)實(shi)際壓(ya)(ya)(ya)(ya)力,而同(tong)一壓(ya)(ya)(ya)(ya)力下(xia)的(de)實(shi)際溫度,含空(kong)氣(qi)(qi)的(de)蒸汽低于飽和蒸汽,這樣導致(zhi)達不(bu)了消毒效果。絕不(bu)可堵塞(sai)安全閥出氣(qi)(qi)孔,必(bi)須留空(kong)位保證(zheng)其暢(chang)通放氣(qi)(qi),發現超溫超壓(ya)(ya)(ya)(ya),立即斷電。

蒸餾器

先(xian)關閉放水(shui)(shui)閥,打(da)開(kai)進(jin)水(shui)(shui)閥,待溢水(shui)(shui)皮管(guan)(guan)有(you)(you)水(shui)(shui)流出(chu),方可接(jie)通電源(yuan),此時可暫時關閉進(jin)水(shui)(shui)閥,待出(chu)水(shui)(shui)皮管(guan)(guan)有(you)(you)蒸餾(liu)水(shui)(shui)流出(chu),打(da)開(kai)進(jin)水(shui)(shui)閥,制備蒸餾(liu)水(shui)(shui)。定(ding)期清洗蒸餾(liu)器內壁(bi),除去(qu)水(shui)(shui)垢,不然(ran)影響(xiang)蒸餾(liu)水(shui)(shui)質量和使用效果。切(qie)不可關閉水(shui)(shui)閥制備蒸餾(liu)水(shui)(shui),否則(ze)燒壞電熱管(guan)(guan)。

(6)鼓風機

定期更換(huan)吸氣(qi)口的過(guo)濾(lv)棉(mian),異常(chang)情況,請電工檢(jian)查。

(7)空調

保(bao)種室(shi)若有(you)生長(chang)藻種,需維持恒定溫(wen)(wen)度(du),反之關閉。一(yi)級(ji),二級(ji)空調視外界氣溫(wen)(wen)調整開關。

二 、衛生消毒

工具衛生消毒

1、濾(lv)水筒式(shi)過濾(lv)器(qi)。筒式(shi)過濾(lv)器(qi)的濾(lv)芯應(ying)用(yong)(yong)一次(ci)更換一次(ci),用(yong)(yong)過的濾(lv)芯浸(jin)泡在1%鹽酸(suan)溶液中消毒12h以上。然后用(yong)(yong)洗(xi)(xi)潔精清洗(xi)(xi),再用(yong)(yong)足量清水沖(chong)洗(xi)(xi)干凈,晾干備用(yong)(yong)。(濾(lv)芯規格應(ying)小于(yu)等于(yu)0.5微米)。

2、試管(guan)(guan)(規(gui)格10毫升(sheng)到20毫升(sheng)),錐形(xing)瓶(ping)(50毫升(sheng)到200毫升(sheng)),500ml錐形(xing)瓶(ping),2.5L瓶(ping)廣口(kou)瓶(ping),移(yi)液管(guan)(guan),移(yi)液槍(qiang)管(guan)(guan)/盒,玻璃氣(qi)(qi)管(guan)(guan),膠氣(qi)(qi)管(guan)(guan),氣(qi)(qi)石(shi),溶劑過(guo)(guo)濾器(qi),培養皿。用過(guo)(guo)后(hou)應(ying)當用清(qing)(qing)水(shui)清(qing)(qing)洗(xi)(xi)(xi)后(hou)浸泡于1% 鹽(yan)酸12h以上(溶劑過(guo)(guo)濾器(qi)除外,浸泡時(shi)相(xiang)近大小器(qi)皿分類(lei)放不同的消毒桶,規(gui)格相(xiang)差大,易導致相(xiang)互碰撞碎(sui)爛)然后(hou)用洗(xi)(xi)(xi)潔(jie)精清(qing)(qing)洗(xi)(xi)(xi)(用毛刷清(qing)(qing)洗(xi)(xi)(xi)試管(guan)(guan)底部(bu)時(shi),不可太用力,否(fou)則(ze)底部(bu)易破裂),再用大量清(qing)(qing)水(shui)沖(chong)洗(xi)(xi)(xi)干(gan)凈,風(feng)干(gan)或曬干(gan),高壓滅菌(jun)后(hou),放入恒溫干(gan)燥箱(膠氣(qi)(qi)管(guan)(guan),氣(qi)(qi)石(shi)除外)干(gan)燥備用。

3、聚乙烯藻(zao)袋(dai)用(yong)于二(er)級藻(zao)類藻(zao)種(zhong)掛袋(dai)培(pei)養或3級擴(kuo)培(pei)(一(yi)般使用(yong)盛裝藻(zao)液12升左右)。先用(yong)清水沖洗(xi)干凈(jing)內外,浸泡(pao)于1% 鹽(yan)酸12h以(yi)上(shang),再用(yong)大量清水沖洗(xi)干凈(jing),風干或曬干,備用(yong)。由于藻(zao)袋(dai)近似密封性,對于防(fang)止微生物污染有很好的效果。

4、毛巾、保種室,一級(ji)和二級(ji)毛巾不可(ke)混用(yong),所用(yong)毛巾后都(dou)應消毒清洗晾干后備用(yong)。

5、錫箔紙:除非(fei)粘(zhan)上油污或污物以及破損,都應重復(fu)使用。

6、牛(niu)皮紙:定期(qi)拿到烘箱烘干。

7、定(ding)期更換空(kong)氣過濾(lv)(lv)器(用于1,2級培(pei)養的進(jin)氣過濾(lv)(lv))的濾(lv)(lv)芯,濾(lv)(lv)芯已(yi)油化(hua)或沖散的棄(qi)去。

8、定期更換鼓風機(ji)進風口的過(guo)濾(lv)棉(mian)。

9、對(dui)新購入的(de)器皿應(ying)先除去(qu)包裝(zhuang)沾(zhan)染的(de)污垢后,用(yong)洗潔精刷洗,流(liu)水沖凈,再(zai)(zai)浸泡于1%的(de)鹽(yan)酸12h,使游離的(de)堿(jian)性物(wu)質除去(qu),再(zai)(zai)用(yong)清水沖洗干凈,風干或曬(shai)干備(bei)用(yong)。

室內衛生消毒

1、保種室每天早上用清水擦洗實驗臺面,地(di)板,墻壁(bi),木架后(hou),開(kai)紫(zi)外(wai)燈(deng)消毒20分鐘,此時(shi)關掉空調,關掉日光燈(deng),由(you)于(yu)紫(zi)外(wai)燈(deng)照(zhao)射會產生一定(ding)量的臭(chou)氧(yang),對人(ren)體有害,所以消毒關閉紫(zi)外(wai)燈(deng)后(hou)人(ren)應隔一段(duan)時(shi)間再進(jin)(jin)去。進(jin)(jin)保種室換鞋。

2、一級和二級室每(mei)天清潔,接種前接種后承載物框架都應(ying)用(yong)消毒液(ye)消毒。

3、門口消(xiao)毒(du)池定期更(geng)換消(xiao)毒(du)藥(yao)水(高錳酸鉀(jia))。

4、常用消(xiao)毒(du)劑濃度 1%鹽酸,次氯酸鈉20ppm(有效(xiao)氯),碘(dian)液(1000ppm),酒精70-75%,高錳酸鉀1mg/L。

三 、接種

藻種固體培養基制備

所需燒(shao)杯(bei),玻棒須清(qing)洗干凈(jing),錐(zhui)形瓶和(he)培(pei)養(yang)皿須消毒(du)清(qing)洗滅菌烘干。將(jiang)稱好的(de)純(chun)化(hua)瓊(qiong)(qiong)脂粉(fen)加(jia)入燒(shao)杯(bei)再(zai)加(jia)入一定(ding)量(liang)的(de)海水(shui),加(jia)熱(必須不停攪拌(ban),以免瓊(qiong)(qiong)脂糊底燒(shao)焦)至完(wan)全融化(hua),加(jia)熱后(hou)(hou)的(de)瓊(qiong)(qiong)脂培(pei)養(yang)基應當透明(ming)無(wu)顯(xian)著(zhu)沉淀,若明(ming)顯(xian),可采(cai)用清(qing)潔的(de)白色(se)薄(bo)絨布或中間夾有(you)薄(bo)層吸水(shui)棉的(de)雙層紗布鋪在漏斗上趁熱過濾(lv),融化(hua)畢后(hou)(hou)精確加(jia)入營養(yang)鹽(yan)(維生素B溶(rong)液須等培(pei)養(yang)基高壓滅菌后(hou)(hou)冷卻至50攝氏度以下再(zai)加(jia))。

培(pei)(pei)養(yang)(yang)基(ji)(ji)采用121攝氏(shi)度(du)高壓(ya)蒸汽(qi)滅菌(jun)20至30分(fen)鐘(zhong)即可(ke),培(pei)(pei)養(yang)(yang)基(ji)(ji)滅菌(jun)后(hou)(hou)應(ying)立即取出,冷(leng)卻至50攝氏(shi)度(du)以下,即加(jia)維(wei)生(sheng)素B溶液(ye)。將盛裝培(pei)(pei)養(yang)(yang)基(ji)(ji)的錐形瓶和培(pei)(pei)養(yang)(yang)皿放在(zai)超凈(jing)工作臺上,點燃(ran)酒精燈,右手托起錐形瓶底,將瓶口(kou)在(zai)酒精燈稍(shao)加(jia)燒,然后(hou)(hou)迅速將培(pei)(pei)養(yang)(yang)基(ji)(ji)倒入培(pei)(pei)養(yang)(yang)皿中(zhong),以鋪滿皿底為止(厚(hou)薄適中(zhong))。鋪放好后(hou)(hou),放置20-30分(fen)鐘(zhong),待培(pei)(pei)養(yang)(yang)基(ji)(ji)凝固后(hou)(hou),即開始操作。

若是做斜面(mian)培養(yang)基,則加入(ru)維生素B溶(rong)液后(hou),將(jiang)盛裝(zhuang)(zhuang)培養(yang)基的錐形瓶和(he)試管以(yi)及一根厚約(yue)(yue)1厘米的小(xiao)木板放(fang)在超凈(jing)工作(zuo)(zuo)臺上。分(fen)裝(zhuang)(zhuang)約(yue)(yue)為試管高度的1/3-2/3(視情(qing)況(kuang)操作(zuo)(zuo)),分(fen)裝(zhuang)(zhuang)量應(ying)以(yi)能(neng)形成2/3底層和(he)1/3斜面(mian)量為好,擺(bai)置成適當(dang)斜面(mian),待(dai)其自(zi)然凝固(gu)。培養(yang)基制備(bei)好后(hou),如發現破裂,水分(fen)浸入(ru),色(se)澤(ze)異常等均應(ying)棄去.

待凝(ning)固(gu)后即進行操(cao)作,現一般采用平板(ban)劃線接種(zhong)和涂布接種(zhong)。

平板劃線接種

涂布接種

所(suo)用種液必須(xu)經過弧菌檢測(ce)合格后方(fang)可使用。涂布(bu)(bu)棒在火焰(yan)上慢慢來回(hui)過火焰(yan)3次,冷(leng)(leng)卻(不可驟(zou)冷(leng)(leng)),將(jiang)種液用移液槍從(cong)試管(guan)吸出注(zhu)入平板(ban)上面(mian)(槍管(guan)不可碰及試管(guan)外(wai)壁,更不能碰及其它(ta)物(wu)品或臺面(mian)),迅速用涂布(bu)(bu)棒在表面(mian)作來回(hui)左(zuo)右的涂布(bu)(bu),讓藻液分布(bu)(bu)均勻,畢后,封口同平板(ban)劃線。

試管液體培養基接種

所用(yong)種(zhong)液必(bi)須(xu)經(jing)過弧菌(jun)檢測(ce)合格后方(fang)可(ke)使(shi)用(yong)。火(huo)焰(yan)滅(mie)(mie)菌(jun)前試管(guan)口(kou)外壁(bi)不(bu)(bu)能(neng)(neng)有水滴(易導致受(shou)熱不(bu)(bu)均而破裂(lie)),燒試管(guan)口(kou)時(shi),試管(guan)傾斜45度角,不(bu)(bu)斷移(yi)動試管(guan),使(shi)試管(guan)口(kou)受(shou)熱均勻(yun),滅(mie)(mie)菌(jun)時(shi)間3秒(miao)左右。滅(mie)(mie)菌(jun)后不(bu)(bu)能(neng)(neng)驟冷,防止試管(guan)破裂(lie)。畢后,用(yong)盛(sheng)裝(zhuang)營(ying)(ying)養液150或200ml的錐(zhui)形(xing)瓶(也(ye)要滅(mie)(mie)菌(jun)瓶口(kou))向試管(guan)中傾倒營(ying)(ying)養液,每次只能(neng)(neng)拿(na)一個試管(guan)(注:若(ruo)幾個試管(guan)對比(bi)時(shi),可(ke)以同(tong)時(shi)拿(na)在(zai)手(shou)里,用(yong)大拇指和手(shou)掌),倒好(hao)后,同(tong)樣試管(guan)和錐(zhui)形(xing)瓶口(kou)滅(mie)(mie)菌(jun),試管(guan)塞也(ye)要滅(mie)(mie)菌(jun),塞試管(guan)塞時(shi)不(bu)(bu)要移(yi)動試管(guan)迎(ying)接塞,以免(mian)試管(guan)在(zai)移(yi)動中納入可(ke)能(neng)(neng)含菌(jun)空(kong)氣,火(huo)焰(yan)旁為(wei)無菌(jun)。

三角瓶,500ml瓶,2.5L瓶液體培養基接種

三角瓶(ping)接種(zhong)(zhong)需(xu)無(wu)菌(jun)操(cao)作。500ml瓶(ping),2.5L瓶(ping)液體培(pei)養(yang)(yang)(yang)基(ji)接種(zhong)(zhong)均(jun)在室內衛生(sheng)條件下(xia)操(cao)作。3種(zhong)(zhong)瓶(ping)和所(suo)需(xu)營養(yang)(yang)(yang)液及錫(xi)紙,移液管,洗耳(er)球,氣(qi)管,氣(qi)石(shi)均(jun)需(xu)經(jing)消(xiao)毒清洗,滅菌(jun)后使(shi)用。三角瓶(ping)需(xu)定時(shi)搖置,500ml,2.5L瓶(ping)培(pei)養(yang)(yang)(yang)在通氣(qi)條件下(xia)培(pei)養(yang)(yang)(yang)。

二級聚乙烯藻袋液體培養基接種

氣(qi)管(guan),氣(qi)石均需經消毒清洗(xi),滅菌后使(shi)用(yong)(yong),抽藻(zao)(zao)管(guan)前次(ci)用(yong)(yong)畢后應(ying)(ying)當(dang)用(yong)(yong)水泵將管(guan)內(nei)灌滿消毒鹽酸(suan)溶液,今次(ci)用(yong)(yong)前需用(yong)(yong)海水或淡水沖洗(xi)干(gan)凈后使(shi)用(yong)(yong)。選取的(de)種液應(ying)(ying)當(dang)是無(wu)明顯沉淀,泡沫,原生(sheng)動物(wu),附著物(wu),色澤正常(chang)(chang),藻(zao)(zao)細胞密度大,形態正常(chang)(chang),弧菌檢測(ce)(ce)合格后選用(yong)(yong)。所(suo)用(yong)(yong)海水漂(piao)白(bai)(bai)水消毒有效氯應(ying)(ying)足10ppm,曝氣(qi)12小(xiao)時,檢測(ce)(ce)余氯,加硫代硫酸(suan)鈉中和,方可(ke)使(shi)用(yong)(yong)。若使(shi)用(yong)(yong)稀釋的(de)漂(piao)白(bai)(bai)水,應(ying)(ying)當(dang)天(tian)配制當(dang)天(tian)使(shi)用(yong)(yong),而(er)未使(shi)用(yong)(yong)的(de)要(yao)(yao)在24小(xiao)時后棄去。且稀釋的(de)漂(piao)白(bai)(bai)水必須加蓋放在避光(guang)的(de)容(rong)器內(nei)保存。由(you)于(yu)漂(piao)白(bai)(bai)水會隨(sui)時間漸(jian)漸(jian)分解,一次(ci)性購入的(de)漂(piao)白(bai)(bai)水不應(ying)(ying)太多,不要(yao)(yao)過(guo)量儲存,以(yi)免影響滅菌功效。
